It is a triple receptor agonist for GLP-1 (Glucagon-like peptide-1), GIP (Gastric inhibitory polypeptide, also known as glucose-dependent insulinotropic polypeptide), and GCGR (glucagon receptor). Retartrutide is a 39-amino acid peptide that is resistant to cleavage by DPP4, the enzyme that cleaves GLP-1 and GIP.
